缺氧对COX-2在胰腺癌PC-3细胞株中表达的影响 被引量:1

摘要 目的 检测环氧合酶-2(COX-2)在胰腺癌细胞株中的表达,并通过二氯化钴(CoCl2)诱发细胞生存的缺氧环境,探讨缺氧对胰腺癌PC-3细胞株COX-2表达的影响。方法 采用免疫组织化学检测COX-2在PC-3细胞株中的表达,运用逆转录-聚合酶链反应(RT-PCR)观测CoCl2与PC-3细胞COX-2表达间的量-效和时-效关系。结果 胰腺癌PC-3细胞株中存在COX-2的表达,CoCl2可上调细胞中COX-2的表达,并且与COX-2表达间表现出一定剂量和时间范围内的依赖性关系。结论胰腺癌PC-3细胞株中存在着COX-2的表达,缺氧可诱导COX-2在细胞中的表达。 Objective To investigate the expression of cyclooxygenase - 2 (COX- 2) and the effects of CoCl2 on the expression of cy-clooxygenase (COX - 2) in pancreatic carcinoma PC - 3 cell lines by inducing Hypoxia. Methods The expression of COX - 2 in pancreatic carcinoma PC- 3 cell lines was studied by immunohistochemistry. The time-concentration relationship and time-effect relationship between CoCl2 and COX - 2 were explored by reverse transcription polymerase chain reaction (RT - PCR). Results COX - 2 was expressed in pancreatic carcinoma PC - 3 cell lines. CoCl2 can up-regulate the expression of COX - 2 in PC - 3 cell lines in a time-dependent and dose-dependent manner. Conclusion COX - 2 was expressed in PC - 3 cell lines, and hypoxia can up-regulate the expression of COX - 2.
