摘要
建立了雷公藤中雷公藤甲素的柱前衍生化毛细管气相色谱分析法,测定了15个不同产地雷公藤去皮根和7种雷公藤根皮中雷公藤甲素的含量。供试品用氯仿提取,酸处理除去生物碱,经重氮甲烷甲酯化,三甲基硅基咪唑衍生化后直接进行气相色谱分析,正三十六烷为内标,线性范围为25—75μg/ml,最低检测量为25ng,相对标准差为9.2%。
A method was developed to determine the content of triptolide in Tripterygium wilfordii from 15 regions of China at levels below those previously achieved by other chromatography. The triptolide was extracted from the powder of T. wilfordii using chloroform, N-(Trimethysily)-imidazol derivatives of the extracts esterified by CH_2N_2 were analyzed by Capillary Gas Chromatography (CGC) in splitless, n-hextriacontane served as the internal standard for quantitation. The triptolide can be easily traced down to the 25 ng level with an overall precision of less than 10% relative standard derivation in the content range of 25—75μg/ml.
