摘要
目的 研究吐温化合物对人胃癌MGC80 3细胞生长的影响 ,确定诱导分化剂二烯丙基二硫 (DADS)的溶媒。方法 采用MTT比色、生长曲线分析、细胞活力检测和流式细胞仪观察溶媒Tween2 0、Tween 80与二烯炳基二硫 (DADS)对MGC80 3细胞生长的影响。结果 Tween 80稀释浓度由 1 0 0 0倍～ 1 60 0 0倍时 ,其他浓度组均具有抑制作用 ,并呈浓度依赖性 (IR 6.2 %～ 92 .6% ,P <0 .0 5 )。Tween2 0稀释浓度由 5 0 0 0倍～ 30 0 0 0倍时 ,均对MGC80 3细胞具有促生长作用 (PR 3.6%～ 1 6.1 % ,P <0 .0 5 )。Tween 80稀释浓度 32 0 0 0倍对MGC80 3细胞生长、生长曲线、细胞群体倍增时间、细胞周期均无影响 ,与对照组、DADS处理组细胞存活率差异无显著性 (P >0 .0 5 ) ,而以此作为溶媒的不同浓度DADS具有抑制MGC80 3细胞生长的效果 ,与对照组和Tween 80组的差异均具有显著性 (P <0 .0 5 ) ,抑制率呈现时间 -效应依赖关系 (P <0 .0 5 ) ,对细胞群体倍增时间延长 (P <0 .0 5 ) ,并且可阻滞MGC80 3细胞于G2 /M期。结论 Tween 80具有抑制MGC80 3细胞生长作用 ,而Tween2 0对MGC80 3细胞具有促生长作用。选择Tween 80稀释浓度 32 0 0 0作为DADS溶媒 ,用于DADS对人胃癌MGC80 3细胞影响的研究比较合适。
Objective To investigate the effects of Tween compounds on human gastric cancer MGC803 cell line in vitro and to ascertain the menstruum of diallyl disulfide. Methods The influence of Tween20, Tween80 and diallyl disulfide were confirmed by MTT, cell growth curve analysis, cell viability detection and flow cytometry on human gastric cancer MGC803 cells in vitro. Results 1 000～16 000 times groups of the dilute concentration of Tween 80 have significant inhibition effect(IR 6.2～92.6%, P<0.05),but 5 000～30 000 times groups of the dilute concentration of Tween 20 have promote effect(PR 3.6～16.1%, P<0.05) for growth of human gastric cancer MGC803 cell in vitro.Tween 80 (32 000 times) had not influence for cell growth, growth curvers, average doubling time and cell cycle, and its cell viability is not significantly,different from that of control group and DADS group(P>0.05). DADS with the menstruum Tween 80 (32 000 times) in vitro inhibited the growth of MGC803 cells and exhibited a dose-dependent model(P<0.05), with average doubling time retarded (P<0.05), and arresting the cell cycle in G2/M phase. Conclusion Tween 80 have inhibition effect and Tween 20 have promote effect for growth of human gastric cancer MGC803 cell in vitro.The menstruum Tween 80 (32 000 times) did not influence cell growth, growth curvers, Average doubling time and cell cycle.
