纳洛酮对缺氧大鼠皮层神经元细胞的保护作用 被引量:15

摘要 目的 观察缺氧对大鼠皮层神经元细胞的影响及纳洛酮的保护作用。方法 取体外培养 12d的Wistar大鼠皮层控经元细胞 ,随机分为正常对照组、缺氧组、纳洛酮组 (缺氧前 2 4h加纳洛酮预处理 ) ;缺氧 6h后在常氧下继续培养 2 4h。观察各种条件下神经元细胞的存活率和形态学的改变 ,测定培养液中乳酸脱氢酶 (LDH)含量。结果 缺氧后可见神经元胞体肿胀、细胞死亡 ,LDH渗出量增多 ,细胞存活率减少 (P <0 0 1)。经纳洛酮预处理的神经元 ,缺氧后神经元胞体肿胀、细胞死亡程度轻于缺氧组 ,LDH渗出显著低于缺氧组 (P <0 0 1) ,而存活率明显高于缺氧组 (P <0 0 1)。结论 缺氧能诱导皮层神经元损伤 。 Objective To observe the damage of hypoxia on the cultured rat cortical neurons ,and to study protective effect of naloxone. Methods Cortical neurons cultured for 12 d were randomly divided into three groups: normal control, hypoxia group and naloxone group (preconditioned with naloxone); the latter two groups were exposed to hypoxic environment for 6 h and then cultured for 24 h under normoxic condition. The variation of survival rate and morphology of neurons were observed, and the contents of lactate dehydrogenase (LDH) in the solution were measured. Results Swelling and cell death occurred after hypoxia treatment, the survival rate of neurons decreased,and efflux of LDH was increased (P<0.01). Compared with the hypoxia group, swelling and cell death were less frequently in naloxone group, lower LDH level and higher survival rate (P<0.01). Conclusion Damage of cultured rat cortical neurons can be induced by hypoxia, which may clearly ameliorate by naloxone.
