酰胺类除草剂与脲酶的相互作用机制研究 被引量:5

摘要 应用紫外差光谱和荧光光谱法研究了水溶液中酰胺类除草剂乙草胺、丙草胺、丁草胺、异丙甲草胺与脲酶分子间的相互作用.结果表明,随着除草剂浓度的增加(0.0~1.6μmol/L),脲酶的紫外吸收光谱发生红移,吸收强度减弱.除草剂对脲酶的荧光均有猝灭作用,且静态猝灭是引起脲酶荧光猝灭的主要原因.从荧光猝灭结果求出除草剂和脲酶的结合常数及结合位点数:乙草胺K=1.17×103L/mol,n=0.81;丙草胺K=1.46×102L/mol,n=0.67;丁草胺K=2.29×101L/mol,n=0.50;异丙甲草胺K=1.49×103L/mol,n=0.84. The binding of acetochor, pretilachor, butachlor and metolachlor to urease in aqueous solution was studied using UV difference spectrum and fluorescence spectrum methods. According to the UV difference spectrum curve obtained, the UV difference adsorption ΔA showed rapid change when herbicide concentrations ranged from 0.0 to 1.6 μmol/L. Red shift occurred in the UV difference spectra of urease and absorbance-peaks were lower. It was shown that the herbicides quenched the urease fluorescence mainly through a static quenching process. The binding constant K and the number of binding site n were calculated according to the fluorescence quenching results. In case of acetochor, (K=1.17×10~3 L/mol) and n was 0.81; to pretilachor, K=1.46×10~2 L/mol and n was 0.67; whereas for butachlor, K=2.29×10~1 L/mol and n was 0.50; and for metolachlor, K=1.49×10~3 L/mol and n was 0.84.
