AT_2受体诱导新生大鼠肾小管上皮细胞凋亡的研究 被引量:3

STUDY OF ANGIOTENSIN II AT_2 RECEPTOR INDUCED RENAL TUBULAR CELLS APOPTOSK

摘要 目的 探讨AT_2受体在诱导肾小管上皮细胞凋亡中的作用。方法 通过取新生大鼠肾小管上皮细胞体外培养,传代后加入不同浓度AT_2受体兴奋剂CGP-42112A,在不同时间用流式细胞仪测定凋亡细胞比值,并结合TUNEL分析。结果CGP-42112A不同浓度加入细胞24h后,随着剂量的增大,凋亡细胞所占比值逐渐增加,与对照组比较均具有显著差异,P<0.001。10^(-5)M CGP-42112A作用于细胞,随着时间的延长,凋亡细胞所占比值逐渐增加,与对照组比较有显著差异,P<0.001。结论 AT_2受体可诱导新生大鼠肾小管上皮细胞凋亡,凋亡细胞数与受体剂量正相关,与受体兴奋剂作用时间正相关。 Objective To investigate the role of Angiotensin II AT2 receptor in renal tubular cells apoptosis. Methods In vitro transfered and cultured renal tubular cells AT2 receptor agonist CGP - 42112A in different doses were diffused, then cell apoptosis was analyzed by TUNEL and flow cytometry. Results With the diffusion of CGP - 42112A in different doses per group respectively (n = 4) , after 24 h, apop-totic cells rations shown difference with the controls, P < 0.001. For CGP - 42112A 10-5 M diffused cells, after 24h, 48h, 60h, 72 h, apoptotic cells were increased compared with control group, P < 0.001. Conclusion AT2 receptor may induce the apoptosis of renal tubular cells. CGP - 42112A has positive dose and time dependent effect on cells apoptosis.
