摘要
采用分析型色谱饼对标准蛋白混合物进行了分离,结果表明装填有小颗粒填料的色谱饼在高流速条件下仍然具有良好的分离能力。在较大流速(5mL/min)条件下,在10min内对人血清白蛋白样品进行了快速纯化,其纯化后的人血清白蛋白的纯度大于85%,回收率为65%,说明分析型色谱饼可以用于快速分离纯化生物大分子。
The purification of human serum albumin (HSA) by using analytical chromatographic pie was developed. Six standard proteins were separated on an analytical chromatographic pie. The results from the resolution of the standard proteins under various flow rates indicate that the chromatographic pie in analytical scale packed with small particles can be utilized under fast flow conditions. The type of mobile phase, concentration of (NH_4)_2SO_4 and pH value for HSA purification were optimized. Crude HSA solution was purified in 10 min in analytical scale with the purity of HSA more than 85% and recovery of 65%. The results show that analytical chromatographic pie is suitable for fast separation and purification of biopolymers.
