摘要 本文报导了人精浆的免疫抑制作用及其机制的研究.我们发现在含有10%混合的 AB^+正常人血清(NHS)同时加入 ConA 和不同浓度 HSP 的条件下培养,人外周血淋巴细胞仍存活,但淋巴细胞转化受到抑制。并且,ConA 活化的淋巴细胞表面 IL—2受体增生受抑制。加入外源性 IL—2能部分地抵消低浓度 HSP 的作用;但是,加入外源性 IL—1对 HSP 的免疫抑制作用无影响。SDS—聚丙烯酰胺凝胶电泳和放射自显影结果表明 HSP 可将^(125)Ⅰ标记的 IL—2至少降解成2个小分子片段。这些结果提示:通过影响 IL—2的产生;降解已产生的 IL—2和减少淋巴细胞表面 IL—2受体的表达可能是 HSP 抑制淋巴细胞转化和增殖的原因之一。这一抑制作用防止了针对精子免疫反应的发生。 Concanavalin A—activated human peripheral blood lymphocytes remained viable for at least 72 hours when exposed to various dilutions of human seminal plasma(HSP)in tissue culture sys-tems containing 10% pooled AB^+ normal human sera,but were inhibited in their ability to un-dergo a blastogenic response(incorporate3~H—Thymidine,3~H-Uridine or3~H—Leucine)or prolif-erate.Activated PBLs cultures containing HSP failed to express high affinity IL—2 receptors suggesting that the IL—2autocrine growth factor mechanism driving T lymphocyte blastogenesis and proliferation was affected by HSP.Electrophoretic analysis of^(125)I—labelled IL—2 revealed degradation(appearance of lower molecular weight forms)after incubation with HSP.Inhibition of lymphocyte blastogenesis by HSP was not affected by the addition of exogenous IL—1 but was partially overcome by addition of exogenous IL—2 These data indicate that HSP degrada- tion the lymphocyte growth factor IL—2,and provide evidence that inhibition of IL—2—in- duced lymphocyte proliferation may be a significant mechanism underlying the immunosup-pressive effect of HSP.
