摘要
本文报告了中国汉族青年学生对PABA乙酰化代谢的表型分布状况。发现64例健康志愿者之PABA乙酰化率均在83.15—97.1%的区间内,属乙酰化快代谢表型,未发现PABA中速或慢速乙酰化个体。对新西兰家兔的研究亦表明它们具有与人类相似的SM_2乙酰化代谢多态性和PABA乙酰化代谢单态性。我们还探讨了新西兰家兔肝匀浆N-乙酰基转移酶对SM_2和PABA的体外乙酰化作用。结果表明同一来源之兔肝匀浆N-乙酰基转移酶对PABA和SM_2两种底物的乙酰化活性互不相同。此外,本文尚建立了产PABA的HPLC检测法。
The distribution of PABA acetylator phenotype in 64 healthy Chinese students wasreported. All the individuals was categorized as rapid acetylator phenotype, in which the percentage ofAc PABA was 83.3-97.1%, with an average of 91.42±3.25% and the variation among individu-als was less than 4%. Neither intermediate nor slow PABA acetylator phenotype was found. showingthat PABA is monomorphic acetylation in Chinese population. Meanwhile, our result also showed thatSM_2 is a polymorphic substrate, and PABA is a monomorphic substrate in New Zealand white rabbitsjust as in human beings. N-acetyltransferase activity of hepatic homogenate of New Zealand whiterabbits of this previously acetylator phenotyped was also determined with SM_2 or PABA as a substratein vitro The finding showed that PABA acetylation activity is not in accordance with SM_2 acetylationactivity in the same homogenate. A HPLC method has been developed for the determination of PABAand its acetylated metabolite in plasma, urine and other biological samples.
