摘要 以磷酸酚二钠为底物在弱碱性条件下经碱性磷酸酶催化水解成苯酚。选择2,6-二氯醌氯亚胺作显色剂,在弱碱性条件下由硫酸铜作催化剂、苯酚反应生成兰色靛酚,在655nm处测定吸光度,与标准系列比较定量可测定各类乳品中碱性磷酸酶活度。检出限为0.02mg/L;标准不确定度0.178~0.246mg/L,可有效判定乳制品巴氏杀菌效果和生鲜牛乳中有机磷酸化合物稳定程度。 Test for ALP activity in a variety of dairy products is based on the principle that ALP hydrolyzes disodium phenol phosphate at weak base, thereby liberating phenol which can be quantitatively detected at 655 nm with phenol standards by color development 2,6- dichloroquinone chloroimide and catalyst copper sulfate. Its limit of detection is 0.02 mg/L and standard uncertainty is from 0.178~ 0.246 mg /L, which can be used for satisfied judging the effect of pasteurization in dairy products and stabilization of organic phosphate compounds in raw milk.
