摘要
在抗肿瘤抗生素(C-1027)产生菌球孢链霉菌(Streptomyces globisporus)中发现一新的质粒pSGL1,其分子量为7.40Kb。 采用Katz等的方法分离提取了质粒pSGL1,pSGL1经单酶切实验证明:PvuⅡ,PstⅠ、XbaⅠ、XhoⅠ和KpnⅠ为单切点;SmnaⅠ为双切点;SalⅠ和BamHⅠ为四切点;而EcoRⅠ、HindⅢ、BgⅢ和PvuⅠ则没有切点,经一系列双酶切实验,构建了质粒pSGLⅠ的限制性内切酶图谱。
A new plasmid pSGL1 was found in antitumor antibiotic C-1027 producing strain Streptomyces globisporus. The molecular weight of this plasmid is 7.40Kb.We have isolated plasmid pSGLl using Katz's method. The experiment of digestion using single restriction endonuclease showed that PvuⅡ, PstⅠ, XbaⅠ, XhoⅠ and KpnⅠ have a unique site respectively in plasmid pSGLl, Sma1 cut the plasmid into 2 fragments, SalⅠ and BamHⅠ cut the plasmid into4fragments respectively,EcoRⅠ,HindⅢ, BgⅢ and PvuI haven't any site in the plasmid. The restriction endonuclease map of plasmid pSGLl has been constructed by a series of double enzyme digestion.
