摘要
目的 探讨正常心肌胶原纤维的分布、及缺血和缺血再灌注对心胶原的影响。方法 成年大鼠 15只 ,分正常组、缺血组和再灌注组。 (1)钳夹冠状动脉左旋支 ,制备缺血组和再灌注组实验动物模型 ;(2 ) 3组切片分别做VanGieson染色和Masson染色。 (3)计算机图像分析系统进行图像分析 ,观察测量各组胶原纤维的含量。结果 (1)每个心肌细胞均有一层胶原鞘 ,心肌细胞群之间有粗大的胶原纤维膜相间隔 ,所有的胶原纤维彼此连续成网。正常心房的胶原纤维含量低于心室各部 (P <0 0 5 ) ;左、右心房之间比较 ;以及左、右心室、室间隔、乳头肌之间比较差异无显著性 (P >0 0 5 )。 (2 )缺血组细胶原纤维有断裂现象 ;粗大的胶原纤维结构松散、凌乱、迂曲 ,内有空泡 ;血管管壁的胶原纤维有破坏。 (3)缺血再灌注组粗、细胶原纤维更加明显断裂、消失 ,胶原网络解体 ,心肌细胞之间的空隙进一步加大。各种微小血管管壁破坏加重。结论 (1)每个心肌细胞和心肌细胞群之间均有胶原纤维膜相包绕 ;(2 )正常心脏各部位胶原的组织学构筑相似 ,但心室含量多于心房 ;(3)心肌缺血可使胶原纤维破坏。
Objective To probe distribution of collagen fibers in normal heart and affect on ischemia reperfusion for collagen of heart Methods Fifteen mature rats were divided into 3 groups, which were normal, ischemic and reperfusion (1)The ischemia reperfusion groups were formed by incarcerated left marginal branch of coronary artery (2)The slices of 3 groupes were used by Van Gieson staining, Masson staining respectively (3) Computer image quantitative analysis was done to observe the amount of separate collagen fibers Results (1) Every cardiac myocyte in the normal heart has a layer of collagen membrane Groups of cardiac myocytes were separated by thick collagen fiber membrane All the collagen fibers were connected into complexed net The amount of collagen fibers in atrium in the normal gruoup was less than that in ventricular parts(P<0 05); The left and right atrium showed no apparent difference(P>0 05)and its also the same case for the comparison between left and right ventric(P>0 05) (2) Some of the thin collagen fibers in the ischemic group are broken, obscued in shape The thick collagen fibers were loosen and in disorder with bubbles inside Some of the collagen fibers in the blood vessel walls were damaged (3)For the reperfusion group, outside the cardiac myocyte collagen fibers breaken and diminished more evidently while the collagen net dismantles and the gap between cardiac myocytes further enlargen Various minor blood vessel walls were damaged more seriously Conclusion (1)Every cardiac myocyte and groups of myocytes have a layer of collagen membrane around them (2)The structures of collagen in every paprt of normal heart are similar and the amount of collagen fibers in atrium is less than that in ventricle (3)The collagen fibers net in the cardiac ischemia are damaged For the reperfusion heart, collalgen fibers were further damaged
