摘要
在油醇和三辛胺混合溶剂为非常规介质的反应系统中,对德氏乳杆菌的乳酸萃取发酵条件进行了研究。结果表明,发酵培养基中的蛋白胨成分,因增加了溶剂在水中的溶解度而对细胞的毒性增大。改良后的培养基B(酵母膏0.5%,牛肉浸膏0.5%,K_2HPO_40.2%,(NH_4)_3C_6H_5O_70.2%,MnSO_4·H_2O0.005%,MgSO_4·H_2O 0.02%,葡萄糖2.5%)宜作为乳酸菌非常规发酵培养时的培养基。研究还表明,对同一浓度的有机溶剂而言,过饱和培养比饱和培养对细胞的毒性更大,而采用固定化细胞培养的方式,能较有效地避免有机溶剂对细胞的毒害作用,并可完成连续操作。
The condition of lactic acid extractive fermentation in the mixture of oleyl alcohol and trioctylamine which was selected as the solvent of extractive fermentation was investigated for the lactobacillus delbruckii. It was found that peptone in medium might improve the solubility of solvent, thus its toxicity to the cells was also increased. The improved medium B (yeast agar 0.5%, beef extract 0.5%, K2HPO4 0.2%, (NH4)3C6H5O7 0.2%, MnSO4·H2O 0.005%, MgSO4·H2O 0.02%, glucose 2.5%) was suitable using in extractive cultivation for non-conventional fermentation of Lactobacillus. The study also indicated that the toxicity of solvent was stronger at the two phase medium than the saturated medium. The method which the cells were immobilized by Ca-alginate gel beads can reduce the toxicity to the cells.
