摘要
实验用HPLC法测定了静脉恒速注射后苦参碱在家兔血浆内的浓度,对10,15,20,30,60mg/kg 5个剂量组进行药代动力学研究。结果表明,苦参碱在家兔体内药代动力学符合二室开放模型,10~30mg/kg时t_1/2β,V_c,CLS基本相似,AUC与剂量成比例增加,呈线性消除。60mg/kg时,t_(1/2)β显著延长,AUC超比例增加,呈非线性消除。
The authors determined the concentration of matrine in rabbit blood plasma with HPLC after it was injected intravenously at constant rate and studied the pharmacokinetics with following five dosage groups of 10,15,20,30 and 60 mg/kg,respectively.As a result,the pharmacokinetics of matrine in rabbit body coincides with two-compartment open model.For 10-30mg/kg dosage groups,the t_(1/2)β,Vc and CLS are similar basically. A direct proportion increase in AUC as dose is increased.The matrine reveals linear elim- ination.For 60 mg/kg dosage group,t_(1/2)β obviously lengthens.AUC increases over pro- portion.The elimination of matrine is nonlinear.
