三七总皂苷对内毒素损伤血管内皮细胞炎症特性的影响 被引量:36

摘要 目的 :研究三七总皂苷对内毒素损伤血管内皮细胞炎症特性的影响。方法 :随机将培养人脐静脉内皮细胞(ECV30 4 )分为对照组、内毒素组、用药组 (内毒素加三七总皂苷 )组 ,各组细胞间粘附分子 1表达用流式细胞仪测定、培养上清中血小板活化因子用生物法测定、培养上清中一氧化氮的变化用Griess法测定。结果 :内毒素加三七总皂苷组与内毒素组相比 ,各指标均较内毒素组有不同程度降低 ,但仍高于对照组。结论 :三七总皂苷对内皮细胞炎症介质产生及粘附分子表达有抑制作用 ,提示其具有抗炎作用。 OBJECTIVE To investigate the effect of total saponins of Panax notoginseng (PNS) on inflammation characters of vascular endothelial cell. METHODS 24 well cell culture platelets on which the vasclular endothelial cell (EVC304) grew were added with endotoxin (ET group). And some of them included PNS(PNS group). After different period of incubation, the expression of ICAM-1 on vascular endothelial cell membrane was assayed by flow cytometry, the content of NO in culture medium was observed by Griess method, the level of PAF in culture medium was assayed by blood platelet aggregation method. RESULTS Comparing with ET group, the expression of ICAM in PNS group reduced at all time point, so did PAF, But NO weren't changed significantly. CONCLUSION PNS can inhibit adhensive characters of endothelial cell by reducing the expression of adhension molecualr and by changing the releasing of some inflammatory media such as PAF.
