摘要
目的 观察 Behcet ) 病患者外周血淋巴细胞 CD80 (B7- 1)、CD86 (B7- 2 )、CD2 8、CTL A- 4分子的表达。 方法 采用流式细胞三色直接免疫荧光标记法 ,对 2 4例 Behcet ) 病患者 (Behcet ) 病组 )和 2 0个正常人 (对照组 )外周血 T、B淋巴细胞亚群的共刺激分子 CD80、 CD86、 CD2 8和 CTL A- 4的表达进行检测。 结果 Behcet ) 病患者外周血 CD4 + T细胞表面 CTL A- 4分子的表达 [(3.18± 1.18) % ]高于对照组[(1.73± 0 .6 6 ) % ],两者差异有显著性的意义 (t=- 3.72 2 ,P<0 .0 1) ;CD19+ B细胞表面 CD86分子的表达 [(4.4 9± 1.73) % ]高于对照组 [(2 .4 0± 1.4 9) % ],两组比较差异有显著性的意义 (t=- 2 .0 71,P<0 .0 5 )。比较两组间其它分子的表达 ,差异均无显著性的意义。 结论 Behcet ) 病患者 B7、 CD2 8的相互作用促进了葡萄膜炎的发生 ,阻断其相互作用可能为今后葡萄膜炎的治疗提供一种新思路。
Objective To investigate the expression of costimulatory molecules( B7, CD28, and CTLA-4) of peripheral blood lymphocytes in patients with Behcet′s ) disease(BD). Methods Lymphocytes were obtained in 24 patients with BD and 20 healthy individuals, and the expression of CD80(B7-1), CD86(B7-2), CD28 and CTLA-4 on T and B cells were detected by direct three-color immunofluorescence flow cytometry. Results Significantly increased expression of CTLA-4 on CD4 + T cells [(3. 18±1. 18)%] was found in BD patients compared with that in controls [(1. 73±0. 66) %] ( t=-3. 722,P <0. 01). The expression of CD86 on CD19 +B cells was also significantly increased in BD patients [(4. 49±1. 73)%] compared with that in controls [(2. 40±1. 49) %] ( t=-2. 071,P <0. 05). No significant difference in the expression of the other costimulatory molecules was found. Conclusions Interaction of B7 and CD28 on peripheral lymphocytes promote the occurrence of uveitis in BD patients. Intervention with these costimulatory signals may lead to a new prevention or treatment for uveitis patients.
