摘要
目的 :探讨多参数流式细胞术分析肿瘤细胞早期凋亡的方法。方法 :采用AnnexinVFITC/PI双染法及JC 1染色法标记食管癌EC6细胞 ,通过多参数流式细胞术分析X射线、顺铂诱导产生的细胞凋亡情况及线粒体膜电位的变化。结果 :随着剂量的增加 ,凋亡细胞的百分率逐渐增加 ,线粒体膜电位下降 ,去极化细胞增多 ,并且有明显的剂量依赖性。结论 :多参数流式细胞术具有广阔的应用前景 ,短时间内能够获得大量统计学意义上可靠的数据 ,为探讨细胞凋亡机制、寻找治疗靶点提供了有益线索。
Objective:To discuss the analysis of cell apoptosis in early stage by multi-parameter flow cytometry. Methods:The apoptosis and the change in mitochondrial membrane potential of esophagus carcinoma cell line EC6, which was treated with X radiation or cisplatin and labeled with Annexin V FITC/PI or JC-1, were analyzed by flow cytometry.Results: The percentage of apoptosis cell and depolarized cell increases with the dose. The mitochondrial membrane potential decreases with the dose. Conclusion: Flow cytometry is a powerful technique in terms of the numbers of cells that can be analyzed and the volume of data generated. It may always be necessary to demonstrate apoptosis and offer potential new targets for therapeutic treatment of diseases.
