摘要
目的 :测定环孢素眼用凝胶的含量。方法 :采用高效液相色谱法 ,YWG C1 8色谱柱 ,流动相甲醇 0 .0 3moL·L 1 磷酸溶液 ( 90∶10 ) ,检测波长 2 10nm。结果 :在 48.8~ 3 90 .4μg·mL 1 范围内 ,环孢素对照品的峰面积 (Y)与其浓度(C)呈良好的线形关系 ,r =0 .9993 ,高、中、低 3种浓度平均回收率 98.7% ,98.9% ,99.2 % (n =5 ) ,RSD分别为 0 .5 1% ,0 .5 6% ,0 .3 7%。平均日内、日间RSD分别为 0 .46% ,0 .5 9%。结论 :该法快速、灵敏、准确。
Objective:To determine the contents of cyclosporin in ophthalmic gel with HPLC. Methods:A reversed-phased high performance liquid chromatographic method was developed. Cyclosporin was chromatographed on YWG-C 18 column, and detected at 210 nm. The mobile phase was methanol-0.030 moL·L -1 phosphate solution (90∶10). Results:The linearity of cyclosporin was over the range of 48.8-390.4 μg·mL -1 with the correlation coefficient 0.999 3. The average recoveries of cyclosporin were 98.7%- 99.2% (n=5) with RSD 0.37%-0.51%. The within-day and between-day RSDs were 0.46% and 0.59% (n=5), respectively. Conclusion:The method is fast, sensitive, accurate and specific for the determination of cyclosporin in ophthalmic gel.
