摘要
目的:检测脑梗死后Fas,Fasl在缺血脑组织中的表达情况,探讨脑梗死的机制。方法:用线拴法制成Wistar大鼠大脑中动脉永久性闭塞模型,精心喂养7d后断头取脑,用免疫组织化学方法检测各脑区Fas和Fasl的表达情况,并与正常组对照。结果:正常脑组织各区有少量细胞表达Fas,梗死侧基底核区、皮质、齿状回和海马表达Fas的阳性细胞数依次增多犤(14.88±0.66),(24.83±0.47),(39.65±1.72),(44.86±1.28)个/高倍视野,F=1933.78,P=0.000犦,对侧基底核区、皮质、海马和齿状回表达Fas的阳性细胞数依次增多犤(13.39±0.93),(23.96±1.25),(33.63±1.22),(92.11±5.09)个/高倍视野,F=1088.13,P=0.000犦,不同部位双侧增高的程度部分不一致,梗死侧海马和基底核区较多(海马F=270.25,齿状回F=36.65,P均=0.000),对侧齿状回较多(F=387.62,P=0.000)。正常脑组织和梗死脑组织基底核区无Fasl的表达,一侧脑梗死后,双侧基底核区仍无Fasl表达,大脑皮质、海马及齿状回均有Fasl的表达,每一侧不同部位表达水不一致(梗死侧F=160,梗死对侧F=61.19,P均=0.000);梗死组不同部位双侧增高的程度部分不一致,双侧齿状回有区别犤(1.00±0.76)比(2.75±0.89)个/高倍视野,t=5.60,P<0.001犦。其中,Fas的表达明显高于Fasl的表达程度。
AIM:To detect the expression of fas and fasl in ischemia brain tissue after cerebral infarction,and explore the mechanism of cerebral infarction. METHODS:Wistar rat models of permanent middle cerebral artery occlusion(MCAO)were made by thread embolism method,and sacreficed for brain after careful feeding for 7 days,immunohistochemistrical method was used to detect the expression of fas and fasl of brain,and contrasted to that of the normal group. RESULTS:Fas expression were found in a few cells in normal brain tissue,number of positive cells expressed fas of basal nuclei in infarction part,cortices,dentate gyrus and hippocampi increased orderly[(14.88±0.66),(24.83±0.47),(39.65±1.72),(44.86±1.28)/high power field,F=1 933.78,P=0.000], those of the other side were[(13.39±0.93),(23.96±1.25),(33.63±1.22),(92.11±5.09)/high power field,F=1 088.13,P=0.000].The bilateral increasing degree was discrepant in different parts,there were more in hippocampi,basal nuclei of the infarction side(hippocampi F=270.25,dentate gyrus F=36.65, P=0.000),and more in dentate gyrus of the other side(F=387.62,P=0.000).There were no fasl expression in normal brain tissue and basal nuclei of infarction brain tissue,there was still no fasl expression in the bilateral basal nuclei after cerebral infarction.There were fasl expressions in cortices,hippocampi and dentate gyrus,and the level of expression was discrepant in different parts of each side(infarction side F=160,the other side F=61.19,P=0.000);The bilateral increasing degree was discrepant in different parts in the infarction group,the bilateral dentate gyrus was different[(1.00±0.76)to(2.75±0.89)/high power field,t=5.60,P< 0.001],and the degree of fas expression was significantly higher than that of fasl. CONCLUSION:High expression of fas and fasl may be one of the mechanism of ischemic brain injury,and the high expression of fasl may play an important role.
