脑缺血再灌流后bFGF基因的表达及MK-801的影响 被引量:6

摘要 目的 研究脑缺血再灌流后 b FGF基因表达的意义及其机制。方法 采用原位杂交和免疫组化的方法 ,观察大鼠脑缺血再灌流后 b FGF基因的表达及 MK- 80 1对它们的影响。结果 缺血 2 h再灌流 1h可见 b FGF表达增高 (P<0 .0 5 ) ,b FGF m RNA表达于 12 h达高峰 ,b FGF蛋白于 2 4h达高峰 ;MK- 80 1组与缺血组相比 ,于再灌流 6 h~ 48h b FGF表达减弱 (P<0 .0 5 )。结论 局灶性脑缺血再灌流可诱导 b FGF表达 ,MK- 80 1则能抑制脑缺血后 b FGF的表达。 Objective To investigate the expression of bFGF and its implications in rat with cerebral ischemia reperfusion. Methods Expression of bFGF was examined by using hibridization in situ and immunochemistry method and on which the effect of a specific uncompetitive N Methyl D A spartate (NMDA) receptorblocker (MK 801),was observed simultaneously. Results bFGF expression increased at 1h reperfusion following ischemia for 2h (P<0.05),bFGF mRNA expression reached peak at 12h,bFGF protein expression reached peak at 24h,The expression of bFGF at 6h~48h reperfusion was decreased in MK 801 treated rats as compared with ischemia reperfusion rats (P<0.05). Conclusions Cerebral ischemia reperfusion can induce the exression of bFGF,which can be suppressed by MK 801.
