摘要
目的:研究再灌注期实施亚低温治疗对大鼠大脑中动脉闭塞(MCAO)后神经元细胞的凋亡及Bcl-2,Bax基因表达的影响。方法:采用Longa线栓法制作MCAO模型,模型成功后分别进行缺血期3h、再灌注期3h、缺血期至再灌注期6h的亚低温治疗。再灌注后24h断头取脑,连续切片作TUNEL染色及Bcl-2,Bax免疫组化染色。结果:①与对照组的Bcl-2阳性细胞百分率犤(19.79±0.92)%犦相比,缺血期亚低温3h组犤(23.04±3.76)%犦和缺血亚低温至再灌亚低温6h组犤(25.47±2.03)%犦的Bcl-2阳性细胞率增加(q=4.19,0.01<P<0.05;q=7.17,P<0.01)。比较单纯再灌亚低温3h组犤(22.21±2.25)%犦与缺血亚低温至再灌亚低温6h组的Bcl-2阳性细胞百分率,前者Bcl-2阳性细胞率较后者低(q=4.11,P<0.01)。②与对照组的Bax阳性细胞百分率犤(52.15±6.18)%犦相比,缺血期亚低温3h组犤(32.35±3.71)%犦和缺血亚低温至再灌亚低温6h组犤(26.86±2.43)%犦的Bax阳性细胞百分率均降低(q=14.21,P<0.01;q=18.95,P<0.01);而单纯再灌亚低温3h组的Bax阳性细胞百分率犤(47.26±4.52)%犦降低不明显(q=2.75,P>0.05)。③凋亡细胞阳性率与Bcl-2/Bax的比值呈负相关关系(r=-0.9759,P<0.01)。结论:①单纯于再灌注期实施亚低温3h不能明显抑制神经元细胞凋亡。
AIM:To investigate the effect of mild hypothermia in reperfusion period on neuronal apoptosis and expression of bcl 2 and bax gene after middle cerebral artery occlusion(MCAO)of rats. METHODS:The MCAO models, made with Longa thread occlusion method,were treated with mild hypothermia for 3 h in ischemia period(group 1),3 h in reperfusion period(group 2),6 h from ischemia to reperfusion period(group 3)respectively.At the 24th hour after reperfusion,the rats were killed and the brains were continuously cut into three sections for TUNEL staining,bcl 2 and bax immunohistochemical staining. RESULTS:①As compared with the control group[(19.79±0.92)%],the percentage of bcl 2 positive cells in group 1[(23.04±3.76)%]and the group 3[(25.47±2.03)%]were significantly increased(q=4.19,0.01< P< 0.05;q=7.17, P< 0.01);the percentage of bcl 2 positive cells in group 2 [(22.21±2.25)%]was lower than that in group 3(q=4.11, P< 0.01).②As compared with the control group[(52.15±6.18)%], the percentage of bax positive cells in group 1[(32.35±3.71)%]and group 3[(26.86±2.43)%]decreased(q=14.21, P< 0.01;q=18.95, P< 0.01),while that in group 2 didn't decreased significantly(q=2.75, P >0.05).③There was a negative correlation between the percentage of apoptosis and the ratio of bcl 2 to bax protein(r=-0.975 9, P< 0.01). CONCLUSION:①Mild hypothermia only in cerebral reperfusion period can not depress neuronal apoptosis and influence the expression of bcl 2 and bax gene significantly, but the effect will be more obvious if mild hypothermia in ischemia period is prolonged to 3 h after reperfusion.②The occurrence of apoptosis can be influenced by the ratio bcl 2 to bax protein.
