摘要
目的 探讨肝细胞水化状态 ,即容积改变时对肝细胞凋亡的影响。方法 动态观察不等渗培养条件下大鼠肝细胞凋亡数 ,[3 H]亮氨酸、[3 H]TdR掺入量 ,及同期培养大鼠肝细胞上清液中ALT、AST、LDH活性。结果 各培养组肝细胞凋亡数随培养时间延长有增高趋势 ,以高渗组较明显 ,于培养 12 0min平均达到了 10 .17个 /每低倍镜视野 ,但与另两组相比无显著差异 (P >0 .0 5 ) ;低渗组肝细胞凋亡增加数与 [3 H]亮氨酸和 [3 H]TdR掺入抑制量之间均无平行关系 ,R值分别为 0 .2 765和 0 .65 68(P >0 .0 5 ) ;高渗组两者间有明显的平行关系 ,R值分别为 0 .93 0 7(P <0 .0 5 )和 0 .9988(P <0 .0 0 1)。培养上清液ALT、AST、LDH活性在正常范围内波动。结论 高渗状态可能诱发肝细胞凋亡 ,但仍是肝细胞对环境变化的生理反应之一。
Objective To explore the effects of hepatocellular hydration state on cell apoptosis in rats. Methods The amount of heapatocellular apoptosis was countered, incorporation rate of [ 3H]leucine into protein and that of [ 3H]TdR into DNA were measured, and the activity of ALT、AST and LDH in the supernatant of cell culture were observed dynamically,as the hepatocytes of rats were cultured under aniso-osmotic conditions. Results In the synchronous cultures of hepatocytes, the number of cell apoptosis was inclined to increase with time going on, which was more marked in the hyperosmotic group (the average of cell apoptosis was 10.17/low power field at 120 minutes), but no difference of statistical significance was found as compared with other groups (P>0.05).The incorporation rates of [ 3H]leucine and [ 3H]TdR were negatively correlated in the hypo-osmotic group(P>0.05), but were highly correlated in the hyper-osmotic group(P>0.05). The activity of ALT、AST and LDH in the supernatant of cell cultures was within normal range. Conclusions The hepatocellular hydration state under the hyper-osmotic condition may promote apoptosis of hapatocytes, but it may be one of the physiological process to modulate hepatocellular metabolism.
