摘要
为提高固相放射免疫法(SPRIA)检测抗-HBs的敏感性和特异性,我们应用固相放射免疫沉淀-聚乙二醇分析法(SPRIPEGA)进行检测抗-HBs的实验研究,并对新生儿接种乙肝疫苗后的抗-HBs应答进行了检查分析.(1)由SPRIA的两步法简化为一步法;(2)以10%以上的胎牛血清封闭可明显降低非特异反应;(3)^(125)I-HBsAg稀释液中含有4%PEG可明显提高方法的敏感性,完全可与Abbott药盒媲美;(4)必要时,可在1h得出结果;(5)用SPRIPEGA检查了161例新生儿接种乙肝疫苗一年内的抗-HBs应答变化,三剂疫苗后20μg和10μg组的抗-HBs应答率分别为100.0%(理论值95.0~100.0%)和97.3±1.9%,GMT分别为1∶448.9±5.9和1∶217.8±8.2.
In order to increase sensitivity and specificity of SPRIA of anti-HBs we have made the studies of a radioimmunoprecipitation-PEG assay (SPRIPEGA) by which anti-HBs responses to hepatitis B vaccine in neonates were tested. 1. Two-step examination of anti-HBs by SPRIA was simplified into one-step by SPRIPEGA; 2. The non-specific reactions were significantly decreased with 10% fetal calf serum added after coating HBsAg;3. The sensitivity was significantly increased in PBS buffer of ^(125)I-HBsAg containing 4% PEG and comparable to that of Abbott Kit (AUSAB); 4. The results of detection can be obtain within an hour; 5. Anti-HBs responses to hepatitis B vaccine in 161 neonates within one year were tested by SPRIPEGA,the seroconversion rates of anti-HBs in neonates after the booster injection of 20μg and 10μg doses of hepatitis B vaccine were 100% and 97.3%,and the geometrie mean titers were 1∶448.9 and 1∶217.8,respectively.
