摘要
检测38例健康人血、11例恶性肿瘤病人血和14例脐带血T淋巴细胞培养4天的上清液对6株瘤细胞增殖的影响.观察到三种血培养上清液的抗癌活力有明显差异,健康人血培养上清液中合有较强的抗癌因子(ACF)活性,它对三株癌细胞(HeLa、MGC-803、AGZY-83a)的增殖抑制明显,对三株白血病细胞(HL-60、K-562、RaJi)的增殖未见抑制.这种ACF的有效成分含量与T淋巴细胞分裂增殖关系密切,经初步分离检测其分子量为67000左右.贮存于4℃冰箱,其活力随贮存时间的延长而逐渐降低,保存一年活力降低约50%.含ACF的上清液对小鼠的淋巴组织与造血组织均有明显触动.文中对健康人血、肿瘤病人血、脐带血培养上清液抗癌活力的差别以及ACF的特性提出了看法.
Blood samples from 38 healthy men,11 cancer patients and 14 umbilical cords were incubated in a PHA medium for 4 days.The culture supernates were tested for their inhibitory effect on growth of 6 tumor cell lines in vitro.It was found that anticancer factor(ACF) activity was present in supernates of PHA stimulated normal T lymphocyte cultures.The ACF activity reached its peak on 4th day.Preliminary characterization demonstrated an active moiety in eluate of gel filtration on Sephacryl S-300 with mol.wt about 67000.The growth of 3 epitheloid-cancer cell lines (HeLa,MGC-803 and AGZY-83a) was significantly inhibited by the supernates containing ACF;but the growth of 3 leukaemia cell lines (HL60,K-562 and RaJi) was not inhibited.The ACF activity of healthy man was significantly higher than that of cancer patient and cord blood.The supernates containing primarily ACF could stimulate lymphocytopoiesis and haemopoiesis in normal LACA mice.The properties and activities of ACF were discussed.
