摘要
文章考察了培养基、碳源、氮源及激素组合对霍山石斛原球茎液体培养生长和多糖合成的影响。在MS、B5、N6、SH及各自1/2基本培养基中,SH最适原球茎生长,1/2MS最适多糖合成,继代培养30d,原球茎增重3.73倍,多糖质量分数为2.78mg/g。蔗糖、葡萄糖和果糖3种碳源中,30g/L蔗糖促进原球茎生长,明显优于其它体积质量的葡萄糖和果糖,提高碳源体积质量抑制原球茎生长促进多糖合成。NH+4与NO-3、NAA与BA或KT的体积质量与比例可以调节悬浮培养中原球茎生长和多糖合成,低体积质量NH+4对生长有利,高体积质量的氮源及ρ(NAA)/ρ(BA)为1∶1时,多糖体积质量较高,原球茎生长和多糖合成呈负相关关系,为优化霍山石斛原球茎液体培养条件提供了依据。
Effects of basal media, carbon-source, nitrogen-source and hormone combinations on the growth and polysaccharides synthesis of protocorm-like bodies (PLBs) of Dendrobium huoshanense were investigated. Among eight basal media including full-and half-strength of MS, B5, N6 and SH, SH was best suitable for the growth of PLBs and 1/2 MS for the synthesis of polysaccharides. After subculturing for 30 d, the fresh weight of PLBs increased 3.73 folds and the polysaccharide content in PLBs was 2.78 mg/g. Sucrose, glucose and fructose were used as carbon sources to test their influences on growth and polysaccharide accumulation in PLBs. Sucrose of 30 g/L markedly promoted PLB growth compared to glucose and fructose. Polysaccharide accumulation in PLBs was enhanced by increasing sugar concentration. The ratios of NH^+4 to NO^-3 and NAA to BA or KT as well as their concentration changes might regulate PLBs growth and polysaccharide synthesis. Lower concentration of NH^+4 was beneficial to PLBs growth,and polysaccharide accumulation in PLBs required higher concentration of inorganic nitrogen or the suitable ratio of NAA to BA (1∶1). The obtained results suggested that there was an adverse relationship between PLB growth and polysccharide synthesis.
