果糖喂养SHR大鼠致胰岛素抵抗与RAAS相关性实验研究 被引量:3

摘要 目的 探讨自发性高血压大鼠 (SHR)给予果糖喂养后所致胰岛素抵抗与肾素 -血管紧张素 -醛固酮系统的相关性。方法 以果糖喂养 10周龄SHR大鼠 8周造成胰岛素抵抗 ,检测收缩压(SBP) ,空腹血糖 (FBG)、空腹胰岛素 (FINS)、肾素活性 (PRA)、血管紧张素Ⅱ (AngⅡ )和醛固酮(Ald)水平 ,分别与对照组对比。结果 果糖喂养组的SBP、FINS、PRA、AngⅡ、Ald均高于对照组SHR ,但胰岛素敏感指数 (ISI)则明显降低 (P <0 0 5 )。AngⅡ与Ald都与胰岛素抵抗 (包括与空腹 ,30分钟 ,6 0分钟胰岛素和胰岛素面积的对数正相关 ,与ISI负相关 )有关 ,且二者与胰岛素抵抗的相关性均有其独立性 ,AngⅡ与IR的相关性更强。 结论 果糖喂养后的SHR大鼠所致胰岛素抵抗与肾素 -血管紧张素 Objective Insulin resistance has been described in patients with essential hypertension. Angiotensin Ⅱ plays a central role in the regulation of systemic arterial pressure. The relationship between insulin resistance and RAAS in hypertension has not been in vestigated in detail. This study is to evaluate relationship between RAAS and IR in fructose fed SHR. Methods Ten week old SHR was randomly divided to receive the breeder's chow (control n =8) or a 60% fructose diet(frutose group n =8), plasma fasting glucose(EFG) and insulin were measured as well the plasma glucose and insulin levels in oral glucose tolerance test (OGTT) (0, 30, 60, 120, 180 min). BP, body weight, renin, aldosteron, angiotensin Ⅱ before and at end of 8 weeks after treatment were examined. Results 1.No difference in body weight were found between the two groups. 2.SBP was higher in fructose group 8 wks after treatment,(224 89±3 18 vs control 207 78±12 51 mmHg, P <0 05) 3 FBG was of no differences between the 2 groups, however, the 30 min, 60 min, 120 min glucose as the area under the curve(AUC) of glucose were significantly greater in fructose group than that in control 4 Fast insulin, 30 min, 60 min, and AUC of insulin were greater in fructose group than that in control with calculated ISI significantly lower 5 Plasma Ang Ⅱ and aldosterone concentration were remarkably higher in fructose group 6 In fructose group, Ang Ⅱ and aldosterone were significantly related to the fast insulin concentration, 30 min, 60 min and AUC of insulin, while negatively related to ISI, with no correlation between renin activity and insulin concentration. Conclusions BP, plasma aldosterone and angiotensin Ⅱ were increased and was associated with significant insulin resistance in fructose fed SHR.
