摘要
摸索和探讨了HPES-1型高级基因转移仪在BALB/C小鼠脾细胞与小鼠骨髓瘤细胞SP2/0电融合中的应用,获得了一组选择参数,采用短脉冲输出方式,脉冲幅度6kV,脉冲宽度50μs,脉冲数2~9,循环数50×2及脉冲休止时间2s的电融合参数,纳合高细胞密度(10~5cell/200μl)增加细胞间聚集的方法,可较容易地获得杂交瘤,并应用该技术成功地制备了分泌人血清白蛋白单克隆抗体的杂交瘤。
The electrofusion with the spleen cells of BALB/c mice and myeloma celis SP2/0 was investigated by using HPES-1 Advanced Gene Transfer System. The optimum parameters were electric field strength of 6 kV, short duration of 50 μs, pulse number of 23, cycle of 50x2 and pulse relaxation time of 2s. The result proved that choosing optimum elcctrofusion parameters and using the method of high cell density (108 cell /200μl) was liable to aquire hybridoma. The specific monoclonal antibodies against human serum albumin have been developed successfully by using this technique.
