红细胞在不同pH值下膜动力学特性与携氧能力的变化 被引量:15

摘要 目的 :研究单个活态人血红细胞在不同pH值 (pH5 .89.0 )下的膜动力学特性与携氧能力 .方法 :用新型快速显微多道分光光度技术和显微生物医学图像分析技术 (fastmulti channelmicro spectrophotometerandimageanalysistech niques,FMIA)对红细胞形态结构、弯曲弹性模量 (KC)、剪切弹性模量 (μ)和吸光度 (A)进行无扰、在位、实时、高灵敏度联合动态连续测量 .结果 :细胞形态随 pH增大而减小 ;KC,μ和形状因子 (roundnessfactor,REF)在pH 7.4 7.6时最小 ,随酸碱度增大而增大 ;A在 pH 7.4最大 ,随酸碱度增大而减小 .结论 :pH值与细胞的形态、膜的变形能力、流动特性和胞内血红蛋白分子氧合能力密切相关 . AIM: To study the dynamic characteristics of membrane and oxygen carrying ability of human single living erythrocyte with different pH values. METHODS: A novel fast multi channel micro spectrophotometer and image analysis techniques (FMIA) were used to investigate the dynamic characteristics of membrane of single intact living human red blood cell (RBC). Multiple parameters such as morphemic structures, bending modulus (K C), shear elasticity (μ), and absorbance ( A ) were simultaneously observed and measured in situ in a continuous, high speed, real time and noninvasive manner. RESULTS: The morphology of the cell became smaller as the pH increased. K C, μ, and roundness factor (REF) was the smallest at the pH 7.4-7.6, and increased as the acidity or the alkalinity increased. Absorbance A was the largest at the pH 7.4, and decreased as the acidity or the alkalinity increased. CONCLUSION: The morphology, the compliance of membrane, the flow character of the cell and the oxygen carrying capacity of hemoglobin molecules inside the cell are closely related to the pH.
