皮瓣移植过程中表皮生长因子的变化及促进组织修复的机制 被引量:4

The distribution change of the epidermal growth factor receptor in the flap transplantation and its mechanism of acceleration of tissue repair

摘要 目的:研究皮瓣移植前后表皮生长因子(EGF)受体分布特点及外源性EGF对受体分布的影响。方法:选择健康Wistar大鼠背部两侧随意皮瓣(2.5cm×1.5cm)模型,于原位缝合后即刻在皮下注入EGF,应用SABC法免疫组化染色。结果:免疫组化阳性着色主要存在于皮肤的表皮、皮下组织和深筋膜中的上皮细胞、成纤维细胞、血管内皮细胞和毛囊汗腺的上皮细胞。EGF受体免疫反应阳性细胞数在皮瓣移植术后1~9d逐渐增加,第9天达高峰(95±6)个,第13天恢复到术前水平(38±4)个。加入外源性EGF后与对照组相比实验组EGF受体免疫反应阳性细胞数在术后1~6d都有明显增加,术后第6天达高峰(128±5)个,高峰期比对照组提前3d。结论:大鼠皮瓣免疫反应阳性细胞主要存在于表皮、皮下组织和深筋膜中的上皮细胞、成纤维细胞、血管内皮细胞和毛囊汗腺的上皮细胞。 AIM:To examine the characteristics of di stribution of the epidermal growth factor receptor(EGF-R)before and after the flap transpl ant ation and the ef-fects of exogenous EGF on the distrib ution of EGF-R.MET HODS:Caudally based random skin flaps(2.5cm ×1.5cm)were raised on the backs of healthy Wistar rats,and EGF was instilled under fla ps after closure. The streptavidin-bi otin peroxidase complex immunohistochemi-cal staining meth od was used.RESULTS:EGF receptors distributed in the epi dermis of the skin,t he vas-cular endothelial cells in the dermi s and hypodermis,the fibroblasts a n d the epithelial cells of the hair follicle and the sweat gland.Compared with p er-i-operation,the quantity of positi ve immunoreactive cells increased g radually from the 1st day to 9th day after opera tion,and reached the peak va lue(95±6)cell犦at 9day,then recovered to the peri-o peration level (38±4)cellsat 13day.Compared with the control g roup,the positive im-munoreactive cells in the trial grou p increased remarkably from the 1st da y to the 6th day after operation and apply ing the exogenous EGF.The peak valu e(128±5)cel犦reached at 6day which was 3days earlier than that of t he control group.EGF-R distributes in the epidermis o f the skin, the vascular endothelial cells in the dermis and h ypodermis,the fibroblasts,a nd the en-dothelial cells and the sweat gland.
