几种海水贝类甲醛浸渍标本DNA的提取及rRNA基因ITS序列的扩增 被引量:11

DNA Extraction from Marine Shellfish Samples Preserved in Formalin and PCR Amplification of the ITS Region of rRNA Genes

摘要 从中国科学院海洋生物标本馆收藏的 8 种甲醛浸渍的海水贝类标本中提取 DNA 并对其 rRNA 编码基因的 ITS 序列进行特异性扩增,在牡蛎 Crassostrea sp.、长肋日月贝 Amussium pleurondectes ( Linnaeus )、扇贝 Volachlamys ringaporinug ( Sowerby ) 与华贵栉孔扇贝 Chlamys nobilis ( Reeve ) 等 4 个物种的标本中成功获得扩增产物。利用图片分析软件得出牡蛎标本的 ITS-1 和 ITS-2 扩增片段分别为 408 bp 和 524 bp,长肋日月贝标本分别 521 bp 和 537 bp,而扇贝 Volachlamys ringaporinug ( Sowerby ) 与华贵栉孔扇贝标本只获得 ITS-1 扩增片段,长度均为 545 bp。 DNA is extracted successfully from some marine shellfish samples preserved in formalin, which are all taken from the Marine Biological Museum, Chinese Academy of Sciences. PCR amplification for the ITS regions of rRNA genes is achieved successfully in the samples of oyster, scallops Amussium pleurondectes (Linnaeus), Volachlamys ringaporinug (Sowerby) and Chlamys nobilis (Reeve). By using the image processing software, information is obtained as below: the lengths of ITS-1 and ITS-2 of the oysters are 408 bp and 524 bp, respectively; the lengths of ITS-1 and ITS-2 of Amussium pleurondectes (Linnaeus) are 521 bp and 537 bp, respectively. We only get ITS-1 amplification products from the samples of Volachlamys ringaporinug (Sowerby) and Chlamys nobilis (Reeve), which are both 545 bp.
