摘要
采用优球蛋白沉淀法和 DNA 沉淀法从人、猪、兔、豚鼠血清中提取 C_(lq)。发现除 DNA法的提取量略低于优球蛋白法外,两法所得 C_(lq)的生物学活性大致相同.所用的4种血清以豚鼠血清中 C_(lq)的含量最高;用间接乳凝试验检测,人 C_(lq)的结合活性最高,豚鼠 C_(lq)次之;用固相 C_(lq)结合-ELISA 法检测,豚鼠 C_(lq)的灵敏度最高,人 C_(lq)的灵敏度最低,这可能是人 C_(lq)中污染了 IgG 所致.说明异种 C_(lq)可以替代人 C_(lq)检测人循环免疫复合物.
The pseudoglobulin precipitation method and the DNA precipitaion method were used for isolating Clq from human,pig,rabbit and guinea pig sera.The biological activi- ties of Clq isolated in both methods were approximately the same,except that its yield was somewhat lower in the DNA method. The amount of Clq isolated from guinea pig serum was bigger than that from each of other sera.For latex agglutination test,human Clq was superior in binding activity,and guinea pig Clq was the next.In solid phase Clq-ELISA for detection of AHG,the sensitivitity of guinea pig Clq was high,while human Clq was poor.This may be due to contamination of human IgG in the isolated Clq.It was suggested that heterologous Clq be used to replace human Clq in the Clq binding test.
