摘要
目的:建立骨髓间质干细胞(mesenchymal stem cells,MSCs)体外培养方法,观察再生障碍性贫血患者(简称再障)MSCs生长状况及临床意义。方法:采集对照组及再障患者骨髓,分离单个核细胞2×105/ml接种于培养皿中,体外培养MSCs,7天后计数MSCs集落数,待MSCs形成融合层后,观察其对造血的支持作用。结果:MSC细胞形态呈长梭形或多边形,体外培养7天后对照组MSCs集落数为(66.96±17.7)/(2×105)单个核细胞,再障患者MSCs集落数为(23±21)/(2×105)单个核细胞,两者有显著性差异(P<0.01)。再障患者骨髓MSCs融合层对健康人骨髓GM-CFU集落生长的刺激作用较健康人骨髓MSC融合层作用明显减弱(P<0.01)。结论:骨髓间质干细胞体外培养可为再障的诊断、疗效观察及发病机制探讨提供一定实验依据。
Objective: To establish a method of isolating and culturing mesenchymal stem cells (MSCs) derived from aplastic anemia(AA) bone marrow in vitro and explore their clinical significance. Methods: The bone marrow was extracted from the posterior superior iliac spine of the aplastic anemia or the controls. The nucleate cells fraction of morrow was enriched based on 1.073 density centrifuge and seeded in cell culture plat by 2 × 105 cells/ml.The clones of MSCs were counted after 7 days cultured. The function of supporting hematopoisis for MSCs was to be observed in single confluence layer after plating. Results: The MSCs had spindle shape, with irregular processes on cell surface. The clones of MSCs in patient with AA were (23 ± 21) / (2 × 105) cells after 7 days cultured and significantly lower than (66.96 ± 17.7)/(2 × 105)cells in control group( P<0.01). The GM-CFU supporting of MSCs in patient with AA at single confluence layer was significantly lower than those in control group(P<0.01). Conclusion: The culturing of AA MSCs in vitro can be served as a clinical indicator of the diagnosis, therapy and nosogenesis of AA.
