单核巨噬细胞及前列腺素E_2在正常人粒系造血调控中的作用

摘要 本文从造血调控的角度探讨了正常人骨髓单核巨噬细胞及其分泌的前列腺素 E_2在粒系造血中的调控效应。前列腺素 E_2是单核巨噬细胞分泌的主要粒系造血负调控因子,呈剂量依赖性抑制 GM—CFU 的生长。10^(-8)M/ml 时抑制效应就很明显,10^(-6)M/ml 时抑制率达50%;消炎痛虽能抑制单核巨噬细胞分泌前列腺素 E,但对体外正常人骨髓 GM—CFU 无明显作用。 The modulation role of monocyte-macrophage and prostagland in E_2(PGE_2)in vitro on grawlopoiesis investigated in normal human by estimating PGE_2 is a major inhibitory factor of monocytc-nacrophage nediated suppression and is found to be a dose depemdent in- hibitor of normal marrow GM-CFU;PGE_2 produce locally within the bone marrow environ- merit or in sites of hematopoiesis could serve as more significant role than that of blood in regulating granulopoiesis;and indomethacin has no direct effect in marrow GM-CFU and did not stimulate GM-CFU in vitro in the absence of monocyte-macrophage especially in appre- ciable numbers.These finding ssuggest that the monocyte-macrophage is implicated as the central cellular element in the control of granulopoiesis.
