摘要
目的 :研究神经节苷脂 GM1 (GM1 )对冷冻处理后的兔同种异体移植神经再生作用的影响。方法 :选择成年大白兔 36只 ,4只作为供体 ,切取其双侧坐骨神经 ,制成 1 0 m m神经段 ,深低温冷冻储存 2周 ;其余 32只随机分为实验组和对照组 ,每组又分为 4个亚组。将兔左侧坐骨神经造成 1 0 mm长缺损 ,用复温后的同种异体神经进行移植桥接。然后 ,实验组局部应用 GM1 1mg/ d,连续 1 4 d,对照组不做任何处理 ,任其自然生长。 4个亚组分别于术后 2、4、8、1 2周进行组织学、电生理、超微结构、形态学定量分析、肌湿重等观察。结果 :两组均无急性排斥反应发生。术后 1 2周 ,实验组和对照组在运动神经传导速度及形态学定量分析方面比较 ,差异有统计学意义 (P<0 .0 5 ) ,实验组优于对照组。结论 :外源性 GM1
Objective:To study the effects of ganglioside GM 1 on the regeneration of peripheral nerve allografts after cryomanagement in rabbits Methods:Thirty six healthy adult New Zealand white rabbits were used in this study Four rabbits were selected as donors. Bilateral sciatic nerves were taken out from the rabbits and cryopreserved in a -86℃ refrigerator for 2 weeks. The other of thirty two rabbits as recipients were divided into two groups (experiment and control) randomly. Each group was divided into four subgroups and 10mm defects was created in left sciatic nerve.The experiment and control of the nerve allograft for bridging the defect of peripheral nerves was done after rapid thawing. The management is as follows: The experiment:local injection of GM 1 (1 mg/d×14).The control: no management. Regeneration of the sciatic nerve for all animals were examined electrophysiologically and hislologically.Result:The acute reject reaction was not found. The statistical difference of nerve conduction velocity(NCV) and image pattern analysis in the two groups was significant in 12 weeks after operation(P<0.05). The regeneration were evident in two groups under the observation of optical and electronic microscrope. Nerve regeneration in the experiment group was superior to that in the control group.Conclusion:The results reveal that GM 1 stimulated the regeneration of allograft peripheral nerves after cryomanagement.
