摘要
目的 阐明哇巴因在原发性高血压血管重塑中的作用。方法 通过MTT法、台盼蓝染色及乳酸脱氢酶活性测定 ,确定不同浓度的哇巴因对ECV 3 0 4细胞株的效应。并通过PCNA和NF κB免疫组化染色加以验证。结果 生理浓度 (0 3~ 0 9nmol/L)的哇巴因能刺激细胞增殖 ,病理浓度 (0 9~ 1 8nmol/L)的哇巴因抑制内皮细胞的增殖 ,引起细胞凋亡。结论 生理浓度的哇巴因参与维持正常血管内皮细胞的增殖 ,其信号转导通路与NF κB有关。病理浓度的哇巴因影响血管内皮的结构及功能 。
Objective To investigate the effect of different concentrat ions of ouabain on endothelial cells. Methods The study was carri ed out on cultured endothelial cells(EVC 304) in vitro. The proliferation of EVC304 was determined by MTT method. The vitality of cells was determined by Trypan blue exclusion and LDH activity. Using immunohistochemistry the proliferating cell nuclear antigen(PCNA) and NF-κB were determued. Results Ouabain stimulate the proliferation of ECV-304 in physiological concentration(0 3-0 9 nmol/L). The optimal concentration of ouabain was 0 3 nmol/L, and the maximal effect time was 1 to 2 hours. The effect of ouabain in pathological concentration(0 9~1.8 nmol/L)inhibit ECV304 proliferation and induce significant apoptosis. Conclusion The physiological concentration of ouabain stimulates proliferation of normal vascular endothelial cells. Ouabain in pathological concentration alter the structure and function of vascular endothelium, suggesting ouabain may involve in the vascular remodeling of hypertension.
