摘要
目的 观察还氧合酶-2(COX-2)选择性抑制剂NS398对人前列腺癌细胞系PC-3的增殖抑制作用,探讨NS398的抗癌机理。方法 逆转录-聚合酶链反应(RT-PCR)用于检测PC-3的COX-2 mRNA的表达情况。采用四甲基偶氮唑蓝(MTT)法观察 NS398对 PC-3的增殖抑制作用。为进一步探讨NS398对PC-3的增殖抑制机制,通过DNA梯形电泳和透射电镜试验对细胞凋亡的情况进行检测。结果 PC-3的 COX-2 mRNA表达阳性。NS398对 PC-3的增殖抑制作用表现为剂量依赖性和时间依赖性。DNA梯形电泳显示特征性的梯形谱带。透射电镜表现为细胞核膜完整,染色质聚集成块、边集,胞浆内可见明显的“空泡”和“凋亡小体”,细胞膜完整,细胞表面的微绒毛消失。结论 NS398对人前列腺癌细胞系PC-3的增殖具有抑制作用,并能诱导PG-3细胞的凋亡。NS398对PG-3的抑制可能是通过诱导细胞凋亡的途径实现的。
Objective To observe the inhibitive effect of cyclooxygenase--2 (COX--2) selective in-
hibitor, NS398, on human prostatic cancer cell lines PG-3 during the proliferative process in vitro and in-
vestigate the anticancer mechanisms. Methods RT-PCR assay was performed to detect the expression of
COX--2mRNA in human prostatic cancer cell lines PC-3. The proliferative inhibition process of PC-3 by
NS398 was observed by MTT assay. In order to investigate the anticancer mechanism of NS398, DNA
ladder assay and transmission electron microscope were employed for the cell apoptosis. Results COX-2
mRNA was expressed in PC-3. The proliferative inhibition exerted by NS398 showed well time-depen-
dent and dose--dependent manner. There were characteristic ladder bands after PC-3 was treated by
NS398 (100μmol/L) for 72 h. Under transmission electron microscopy, the membrane of cell nuclear
was intact, and chromatin congregated into mass and collected along side. The 'vacuole' presented in the
cell plasm and apoptosis body was observed. The membrane of cell was whole and the microvilli on it was
disappeared. Conclusion COX--2 is closely correlated with the tumorogenesis of prostatic cancer. NS398
can inhibit the proliferation of the prostatic cancer cen lines PG-3 and induce apoptosis of them. These
findings suggest that the inhibitive effect of NS398 on PG-3 is probably achieved by the way of cell apop-
