摘要
目的 探讨IL - 1α对体外兔软骨细胞增殖及胶原基因表达的影响。方法 在成功分离培养软骨细胞后 ,以MTT法检测软骨细胞活性并绘制其生长曲线图 ;将浓度为 1× 1 0 4U/L、1× 1 0 5U/L、1× 1 0 6U/L的IL - 1α作用于贴壁后的软骨细胞 ,以原位杂交方法检测软骨细胞Ⅰ、Ⅱ型胶原mRNA表达情况。结果 IL - 1α可促使软骨细胞向成纤维样细胞形态转化 ,抑制软骨细胞分裂、增殖 ,诱导非特异性Ⅰ型胶原表达 ,抑制其特异性Ⅱ型胶原表达。结论 IL -
Objective To study the effect of IL-1α on chondrocyte proliferation and collagen gene expression. Methods the activity of chondrocyte was detected by MTT method and its growth diagram of curves was drawn after the rabbit chondrocytes being cultured succssfuly,the effects of 1×10 4U/L,1×10 5U/L as well as 1×10 6U/L IL-1α and the hybridization expression of collagen ⅠandⅡin situ are detected also. Results IL-1α could restrain chondrocyte from prolixferation ,transform it into similar fiber cell, induce the expression of non-specific collagen Ⅰand restrain the expression of specific collagen Ⅱ. Conclusion IL-1α is a important factor chondrocyte differentiation.
