摘要
重组质粒pCG25(IFN-γ)转化大肠杆菌DH5α,接种LB培养基,在32℃培养,42℃诱导后经SDS-PAGE凝胶电泳分析,γ-干扰素基因在大肠杆菌细胞中表达的蛋白质为18Kd,诱导6h达到最高峰,表达活性为1 2×108IU/L菌液,对酸(pH2)和热(60℃)不稳定。
The plasmid(pCG25)containing human interferon gamma gene was transformed into E.coil DH5a,The interferon proteins were highly expressed in E.coli DH5a after temperature induction.The expression protein was 18?kd by SDS-PAGE analysis.The average antiviral titer of the expressed product was about 4.1×10~7~1.2×10~8 IU per liter culture and unstable in the conditions of pH_2 and 60℃.The results showed that the expression protein was interferon gamma.
