摘要
本文采用核苷酸指导下的定位突变方法,以Asp置换了人α型干扰素86位的Cys,构建成突变体IFN-αI(86D)。后者在大肠杆菌中的表达水平比其母株(IFN-αI/86C)提高了2.8倍,IFN-αI(86D)在4℃的稳定性也较母株为高。
An interferon mutant, IFN-αI ( 86D ) ,was constructed by substitution of Cys at position 86 with Asp using nucleotide-directed site-spscific muta-genesis method. The IFN-αI ( 86D ) mutant was shown to be more stable at 4℃ as compared with its parent strain, IFN-αI ( 86D ) . The expression lerel of the mutant gene in E.coli was 2.8 times higher than that of its parent gene.
