摘要
目的 筛选出最佳随机引物并优化其反应体系用于 8种细菌的随机引物聚合酶链反应 (AP PCR)分析。方法 对 2 0 0条随机引物进行过筛 ,应用反应曲面设计 (RSD)进行AP PCR反应体系的优化。结果 8种细菌各有 1条最佳随机引物 ,相应的优化反应体系 (关键反应成分 )浓度差别较大 ,其中模板浓度差别最大达 10 0倍。结论 以最佳随机引物及优化反应体系 ,才可使AP PCR分型条件“标准”化。
OBJECTIVE To screen the primers and optimize the reaction systems for arbitrarily primed polymerase chain reaction(AP-PCR) on 8 sorts of bacteria. METHODS A total of 200 kinds of primers were screened and the optimum reaction systems of AP-PCR were obtained by reaction surface design (RSD). RESULTS One kind of optimum random primer was found in every of 8 sorts of bacteria. The differences were observed among the concentrations of optimum reaction system (key reaction components) and the greatest difference between the template concentration was up to 100 times. CONCLUSIONS To standard the condition of genotyping, the best random primer and optimum reaction system must be considered.
