摘要
目的 :探讨肠复康合剂对裸鼠移植性人结肠癌 HT- 2 9血管生成的机制。方法 :将 2 4只裸小鼠随机分为模型组、肠复康组及西药组 ,每组 8只。各组分别于建立人结肠癌 HT- 2 9癌细胞株裸小鼠皮下移植瘤模型后 6 d给予相应药物 ,免疫组化染色结合图像分析系统半定量检测移植瘤组织微血管密度 (MVD)、血管内皮生长因子(VEGF)及其受体 (FL K- 1)、内皮抑素 (ES)的积分光密度 (IOD) ,计算 VEGF/ES比值 ,并使用逐步引入剔出模型进行单因素和多因素线性回归分析。结果 :与模型组相比 ,肠复康组可使移植瘤 MVD、VEGF、FL K- 1及 VEGF/ES比值显著降低 (P <0 .0 1) ,同时使 ES含量明显增高 (P <0 .0 1) ;西药组可明显降低瘤组织 VEGF、FL K- 1含量 (P<0 .0 1) ,但不能使移植瘤 MVD、ES和 VEGF/ES比值减少 (P >0 .0 5 )。单因素回归分析显示 ,瘤组织 VEGF、ES和 VEGF/ES比值与移植瘤 MVD相关 ,多因素回归分析显示 ,移植瘤 MVD与 VEGF/ES比值呈明显正相关 (r=0 .76 2 ,P <0 .0 1)。结论 :肠复康合剂能抑制人结肠癌 HT- 2 9裸鼠移植瘤血管生成 ,其机制可能是降低瘤组织VEGF/ES比值。
Objective: To investigate the mechanisms of Changfukang Mixture(CM) on angiogenesis of transplanting tumor tissues caused by human colon cancer HT-29 cells in nude mice. Methods: After the nude mice model of the subcutaneous transplanting tumor was established by human colon cancer HT-29,24 cases were randomly divided into model group,CM group and western medicine group( n =8,each),treated with saline,CM and western medicine respectively. The tumor tissues were stained by immunohistochemical methods. Microvascular density(MVD) and integral optical density(IOD) of vascular endothelial growth factor(VEGF),fetal liver kinase-1(FLK-1) and endostatin(ES) in the tumor tissues and the cells and the interstitial tissues were semiquantified by image analysis system,and the ratio of VEGF and ES was calculated. Binary and multiple linear regression was adopted to analyze the results with stepwise method.Results: In comparison with that in the model group,MVD in the tumor tissues was significantly reduced ( P <0.01) with the contents of the VEGF decreased in the tumor cells,that of FLK-1 and the ratio of VEGF and ES decreased in 四川省自然科学基金资助项目(No:01SY051-35) the tumor interstitial tissues in CM group( P <0.01) ;In the western medicine group,the contents of the VEGF and the FLK-1 in the tumor tissues were significantly reduced ( P <0.01),but MVD and the contents of ES and the ratio of the VEGF and ES were not significantly reduced ( P >0.05). In addition,MVD was highly positively correlated with the content of the VEGF and the ratio of VEGF and ES and was negatively related to ES in the tumor tissues by single factor analysis. MVD was significantly positively correlated with the ratio of VEGF and ES in the tumor tissues by multiple factor analysis ( r =0.762,P <0.01). Conclusion: CM could inhibit the angiogenesis of the transplanting tumor tissues in nude mice caused by human colon cancer HT-29. Its mechanisms may be related to reducing the ratio of VEGF and ES in the transplanting tumor tissues.
