健脾药对免疫介导再生障碍性贫血小鼠IL-3、IFN-γ影响的实验研究 被引量:7

An Experimental Study of Spleen - Invigorating Drugs on the Influence of IL - 3 and IFN - γ on Immunoin terventing Aplas-tic Anemia Mice

摘要 目的:检测免疫介导再生障碍性贫血(简称再障)小鼠骨髓造血细胞在含健脾药血清的液体培养体系中IL-3、IFN-γ的浓度,探讨健脾药治疗再障的作用机理。方法:用健脾药灌喂SD大鼠,抽取、制备含中药血清;Balb/C小鼠经60Co5.5Gyγ射线照射后由尾静脉输入取自DBA/2小鼠胸腺淋巴结混合细胞悬液形成免疫介导再障模型。在含健脾药血清的液体培养体系中培养再障小鼠骨髓造血细胞,检测培养体系中IL-3、IFN-γ的浓度。结果:健脾药对再障小鼠IL-3、IFN-γ的总量无明显影响。结论:健脾药可能通过使局部IFN-γ浓度降低进而影响造血干/祖细胞增殖、分化。 Objective: The concentration of IL - 3 and IFN -γ of immunointerventing aplasvic anemia mice bone marrow hematopoietic cells in the culture system of spleen - invigorating drugs liquid (SDL) was determined and to explore the mechamism of SDL for aplastic anemia (AA). Methods: Feeding the SDL to rats, withdrew and prepared SDL serum; the Balb/c mice were radiation by 60 Co 5. 5 Gy γ and input the mixed cellular suspension of thymicolymphnode of DBA/2 mice to the coccygeal vein to establish the immunointerventing AA model. The bone marrow hematopoietic cells of mice in SDL system were cultured and the concentration of IL - 3 and IFN -γ were determined. Results: SDL showed no definite influence on the total content of IL - 3 and IFN γ in AA mice. Conclusion: SDL may exert on the decrease of local concentration of IFN -γ to affect the proliferation and differentiation of hematopoietic stem cells and progenitor cells.
