摘要
目的探讨扶正养营颗粒对免疫介导再障小鼠的作用机制。方法BALB/c小鼠40只随机分为正常组、模型对照组、环孢霉素A对照组、扶正养营汤实验组,复制免疫介导再生障碍性贫血小鼠模型,用TUNEL及免疫组化法检测各组小鼠造血细胞凋亡以及相关调控蛋白Fas与Caspase-3的表达。结果与正常组比较,模型组小鼠骨髓造血细胞凋亡率明显升高,Fas与Caspase-3蛋白表达明显升高,差异有显著性(P<0.05);与模型组比较,环孢霉素A组、扶正养营颗粒组小鼠骨髓造血细胞凋亡率明显降低,Fas、Caspase-3蛋白表达明显减少,差异有显著性(P<0.05)。结论扶正养营汤可能通过下调Fas及Caspase-3基因蛋白的表达,延缓造血细胞凋亡,发挥其治疗再障的作用。
Objective: To explore the mechanism of Fuzhengyangyinggranules (FZYYG) in treating immune meditated aplastic anemia(AA). Methods: Forty BALB/c mice were randomly allocated to four groups: normal control group, model group, CSA group and FZYYG group. Animal model of immune meditated aplastic anemia was built up, model group was fed with physical salts, CAS and FZYYG groups were fed with CSA and FZYYG respectively, TUNEL and immue histo-chemistry methods were used to demonstrate the haematogenous apoptosis and gene protein expression of Fas and Caspase-3. Results: Apoptosis rate and Fas and Caspase-3 positive rate in model group were higher than that of the normal group (P<0.05). But the Apoptosis rate and Fas and Caspase-3 positive rate in the two treated groups was significantly lower than that of the model group (P<0.05). Conclusion: FZYYG could suppresse Fas antigen expression and Caspase-3 antigen expression to postpone haematogenous cells apoptosis.
