摘要
目的 :建立高效液相色谱法测定人血浆中麦考酚酸的药物浓度。方法 :色谱柱为SupelcosilTM LC18( 5 μm ,4 .6mm×15 0mm) ;流动相为甲醇 0 .0 2 5moL·L-1磷酸二氢钠溶液 (含 1%的醋酸 ,pH5 .0 ) ( 4 5∶5 5 ) ;柱温 :2 5℃ ;流速为 1.2mL·min-1;检测波长为 2 18nm。结果 :本测定方法的线性范围 :0 .2 5~ 5 0mg·L-1,r =0 .9998。平均回收率为 ( 10 2 .4± 5 .8) % ,日内RSD≤ 7.0 % ,日间RSD≤ 7.9%。结论 :本方法简单、灵敏、重现性好 ,适用于临床血药浓度监测 ,以及人体药动学的研究。
OBJECTIVE To establish a method for determination of mycophenolic acid (MPA) in plasma by HPLC.METHODS A supelcosil TM LC 18 column(5 μm, 4.6 mm×150 mm)was adopted. Mobile phase was a mixture of methanol and 0.025 mol·L -1 NaH 2PO 4 (containing 1% acetic acid, pH 5.0 )(45∶55) .The temperature was set at 25℃ and the flow rate was 1.2 mL·min -1 . The UV detector was operated at 218 nm.RESULTS The assay procedure was shown to produce linear calibration curves over the range of 0.25 to 50 mg·L -1 of MPA in plasma (r= 0.9998 ). Within the range, the recovery rate was ( 102.4 ± 5.8 )%. And the relative standard deviation (RSD) within a day and between days were less than 7.0% and 7.9% , respectively. CONCLUSIONS This method is found to be reproducible, convenient and sensitive for determination of plasma concentration and clinical pharmacokinetic study of mycophenolic acid.
