摘要
利用柞蚕核型多角体病毒(AntheraeapernyiNuclearPolyhedrosisVirus,ApNPV)作为载体研究了报告基因(β-半乳糖苷酶基因)在柞蚕幼虫体内的表达。结果表明 :重组ApNPV能够使柞蚕幼虫产生典型的脓病病症 ,但不形成多角体。柞蚕核型多角体病毒作为表达载体能够使β-半乳糖苷酶基因在柞蚕幼虫体内得到高效表达。雌性幼虫个体的表达量为2.4×107u,雄性幼虫个体的表达量为1.8×107U;雌性个体较雄性个体在表达时相上早96h;对于同一个体 ,组织细胞中β-半乳糖苷酶的含量较血淋巴上清液中β-半乳糖苷酶的含量高。SDS-PAGE结果显示,所表达的β-半乳糖苷酶分子量为116kD。
The expression of report gene(β-galactosidase gene)was studied by using recombinant Antheraea pernyi nuclear polyheˉdrosis virus(rApNPV).The results showed that rApNPV can develop typital symptoms as cloned ApNPV in tussah,but no polyhedra formed.rApNPV can makeβ-galactosidase gene be correct expressed at high level.The total expression level ofβ-galactosidase proteins reached2.4×10 7 U per female larva and1.8×10 7 U per male larva.In the same larva,the expression level is higher in the tissue cells than in the haemolymph supernate.The SDS-PAGE result showed the molecular weight ofβ-galactosidase expressed is116kD.It was the first report of using ApNPV expression vector express foreign gene in tussah larvae.
