摘要
目的 :观察建立慢性阻塞性肺疾病 (COPD)大鼠模型时呼吸道损伤的动态变化过程。方法 :通过气管内注入脂多糖加香烟烟熏法建立COPD大鼠模型 ;观察大鼠气管和肺组织的病理变化 ;检测大鼠血清和支气管肺泡灌洗液 (BALF)中白细胞介素 8(IL 8)的含量。结果 :大鼠气管纤毛受损 ,肺泡周围炎症细胞浸润程度逐渐加重 ,黏膜下腺体肥厚 ;肺泡结构破坏、互相融合形成肺大泡 ;肺泡腔内巨噬细胞溶酶体增多 ,线粒体肿胀 ;随着造模时间的延长 ,大鼠平均肺内衬间隔逐渐增加 ;平均肺泡数逐渐下降 ;血清和BALF中的IL 8逐渐升高。结论 :大鼠气道内注射脂多糖加烟熏的复合刺激法能够成功地建立COPD大鼠模型 ;
Objective To observe dynamical changes of injured airway by way of establishing rat models of chronic obstructive pulmonary disease (COPD).Methods The rats were intratracheally instilled with lipopolysaccha- ride(LPS) twice and exposed to cigarette smoke to make COPD models,whose morphological changes of airways and lung tissue were dynamically observed; interleukin-8(IL-8) in the serum and bronchoalveolar lavage fluid (BALF) were detected. Results After the model are established,the epithelial cilia cohered,partially fell off;the amount of macrophage lysosome in pulmonary alveolus lumen increased and emphysema was noted;mean lung internal lining interval(MLI)significantly increased, mean alveolar number(MAN) decreased significantly, IL-8 in serum and BALF increased significantly.Conclusion COPD rat models are successfully established by passive cigarette smoking plus intratracheal instillation of LPS,and their degree of inflammation in the airway is correlated with its morphological change.
