摘要
利用毛细管区带电泳(CZE)技术探讨了分离小麦子粒醇溶蛋白的最佳条件。用磷酸缓冲液(0.1mol/L磷酸,pH2.5,含22%乙腈(ACN)和1%聚乙二醇20000(PEG20000)),在毛细管长度46.5cm,内径75μm,运行电压15kV,毛细管温度25℃的条件下,可获得高分离度和高重现性的电泳图谱,一个品种在50min内可分离出40个以上的醇溶蛋白组分。利用该条件鉴定了5个小麦品种。
The optimal conditions of gliadin capillary zone electrophoresis (CZE) were studied in this paper. High resolution and reproducibility of gliadin electrophoregrams were achieved by using 46.5 cm length (75 μm) capillary, 15 kV at 25℃ in 0.1 mol/L phosphoric acid buffer( pH 2.5), containing 22% acetonitrile and 1% PEG20000. Using the conditions, one cultivar can be separated into more than 40 gliadin components in 50 min. Five cultivars were identified based on the conditions.
