摘要
利用毛细管区带电泳(CZE)检测了在不同电泳条件下分离小麦种子醇溶蛋白的效果。结果显示,用碱性缓冲液(0.06mol/L硼酸钠,pH9.0,并含有20%乙腈和1%SDS)及47cm×50μmi.d.毛细管柱,在15kV电压和30℃的条件下可获得较高的分辨度和重复性。
Gliadins are major storage proteins of wheat endosperm. They are very heterogeneous and complex in composition, requiring powerful techniques for analysis. By using capillary zone electrophoresis (CZE) the different conditions, including buffer, capillary length, and diameter, temperature and voltage etc., were studied in this article to improve the resolution and reproducibility of gliadin separation. The results showed that excellent resolution and reproducibility were obtained by using the following CZE conditions: basic buffer was 0.06mol/L sodium borate (pH 9.0), containing 20% acetonitrile (ACN) and 1% SDS. Capillary length was 47cm (50μm i.d.). CZE was performed on a Beckman P/ACE 5500 system at 15kV and 30℃. Thus one cultivar can be separated into more than 40 components of gliadins. This resolution is much higher than that of acid polyacrylamide gel electrophoresis (A PAGE). Furthermore, peak migration time varied by a maximum of 0.03min.
