丙烯醛固定,PAP与胶体金免疫电镜双标记技术在神经细胞内抗原标记的应用 被引量:1

摘要 本实验采用了PAP和胶体金免疫电镜双标记技术,对下丘脑室旁核神经元內进行标记,证实了室旁核内有血管加压素与生长抑素共存的神经元.为了加强抗原的固定效果,提高免疫反应的敏感性,我们采用了丙烯醛固定的方法,对比了浸泡固定和灌流固定的效果,在电镜水平证实了丙烯醛浸泡固定同灌流固定一样,均能获得良好的神经组织的超微结构和阳性反应。同时,我们分别采用了包埋前PAP与胶体金双标记和包埋前PAP结合包埋后胶体金双标记两种方法。结果表明,包埋后胶体金染色比包埋前较易发现阳性标记。 Immunoelectromieroscopic doublc labeling technique with PAP and colloid gold was used in present study to label neurons of the hypothalamus paraventricular nucleus. We demonstrated the co-existence of the vasopressin and the somtostatin in paravontricular nucleus. Acrolcin was used in present study in immersing way to improve fixativc effect of the antigens and increase the sensitivity of the immunorcactivity. The results showed that fixative effect of the acrolein in immersing way was the same as that in perfusing way. We obtained well ultrastructure and positive reactivity of the nervous tissue. We used preembedding PAP-colloid gold and preembedding PAP- poste mbedding colloid gold double labelling methods. The results showed that postembedding colloid gold stain demonstrates the positive labellings easier than preembedding colloid gold stain.
