成年wistar雌鼠17只。随机分为对照和实验组。实验组喂服醋酸棉酚(100mg/kg/d)一月后交配。受精后第10天剖腹,观察妊娠动物数与着床胎数,并分别对卵巢组织化学(△~5-3β-羟甾脱氢酶)观察以及和心、肝、子宫等作组织学方法观察。发现醋酸棉酚对大鼠具有明显抗着床作用,抗着床率为100%。此时,阴道涂片显示动情周期紊乱;黄体细胞的△~5-3β-羟甾脱氢酶活性明显下降,子宫内膜相当于间期时的内膜。以上结果提示△~5-3β羟甾脱氢酶活性下降,孕酮合成受阻,可能是醋酸棉酚抗着床作用的机理之一。
17 mature wistar female rats ,7 were orally administered with gossypol acetic acid suspension at 100mg/kg/day, 5 days a week for 30 days. While 10 were given the same amount of distilled water as control. Vaginal smear was taken daily. After 1 month of treatment with gossypol, they were mated. Inhibition of implantation (100%) was observed, in the gossypol-treated rats, the estrous cycle was stopped; Δ~6-3β-hydroxysteroid dehydrogenase activity was reduced; the endometrium was remained, in the diestrus period. A possible mechanism of anti-implantation effect of gossypol seems to be due to the decrease of Δ~5-3β-SDH activity and blocking of the biological synthesis of progesterone.
